E-mu first made two special 16Mb ROMs for those slots: one contained the E-Synth set of sounds (came standard in the E-Synth models of the E4 line), and the Orbit/Phatt ROM containing the sounds from the Planet Phatt and Orbit v1 Proteus (second generation) sound modules.

Later, E-mu introduced the Proteus 2000 series modules (third generation, but who's counting), and designed the sound ROMs from those units to be readable in the E4 samplers (well, I'm not sure about the very first series E4s, but I'm pretty certain they'll run in any of the Classic or Ultra series units). Some of these were 16Mb, but most were a full 32Mb of sounds.

Now, as far as I know, they presets that the sampler would read to access those sounds aren't on the ROMs themselves. The original E4 ROMs came with a diskette that contained the presets, and had to be copied into the samplers internal preset Flash RAM before they could be used. The P2K ROMs didn't give you any E4 presets, but the E4 would read the P2K instruments as samples or presets (sorry, haven't actually tried this personally with the ROMs I have, so I can't exactly confirm.... but I do know that you don't get access to the P2K presets in the E4).
