Yes to both.
To resample from the inputs while playing:
From Preset edit, go to New Sample (Assignable Key 2 in 4.1 or earlier) From New Sample, set Source to Analog inputs. Now, you're set to sample - depending on which mode you want to use (Key Arm or Force), you can start sampling when a specific MIDI key is triggered, when the input level reaches a certain threshold, or just when you press the Force key. If you've got a lot of tracks going, Key would be tricky, unless you were sure the sound you wanted to sample was the ONLY sound that was on that MIDI key. Make sure monitor is off, or you'll get a feedback loop.
To resample internally, do the above, but set the Source to Internal. Use 18 or 20-bit if you find yourself clipping and need more headroom. I've actually never tried to resample while manually twiddling a knob, but it should work. If it doesn't just record the knob movements into a sequencer, then playback from that and resample internally. Keep in mind that internal resample only resamples the signal that is present at the Main outs, so you may have to do some soloing and rerouting of voices to resample this way.
